Mechanism of Action of Acitretin |
Acitretin is a second generation retinoid. It slows down the proliferation of the skin cells and is therefore used in psoriasis. Retinoic acid activates Retinoic acid receptors (RARs) and retinoic acid X receptors (RXRs) and produce effects on gene expression. Retinoid binds to transcription factors (nuclear receptors) and form complex. This complex binds to promoter regions of target genes and regulates their expressions. The gene product formed produce effective pharmacological actions. |

Typical Dosage for Acitretin |
Adult: 25- 30mg / day for 14 – 28 days, then adjusted according to response. Maintenance dose: 25 – 50mg/day. Maximum dose: 75mg/day Darier`s disease: 10mg/day Children: 0.5 mg/kg daily dose can be increased to 1mg/kg daily. |

Effects of Missed Dosage of Acitretin |
Take the missed dose as soon as noticed and if it is the time to next dose then skip the missed dose. Continue the regular dose. Do not double the dose. |
